What causes us to laugh biologically speaking?

I don't know if you mean evolutionarily speaking or chemically speaking. I think you are speaking about the latter. Laughing is caused when your brain receives a stimulus and in return fires hormones like serotonin, and dopamine. This causes the happy and pleasurable feeling when you laugh.
